How Colour Vision Differences Are Detected

Colour vision variation comes from the pigments in retinal cone cells, and the tests used to detect it are built to exploit the specific confusions those pigments produce.

Colour vision differences affect a noticeable minority of men and far fewer women. The cause sits in the retina, and the tests for it are designed around exactly how the difference works.

Colour comes from three overlapping sensitivities

The retina contains cone cells carrying pigments that respond most strongly to short, medium and long wavelengths of light, roughly corresponding to blue, green and red regions.

No cone detects a colour by itself. The brain compares the relative responses of the three types, and colour is reconstructed from those ratios.

This is why a difference in one pigment does not remove a colour but collapses distinctions, since two wavelengths that previously produced different ratios now produce similar ones.

The genetics explain the sex difference

The genes for the medium and long wavelength pigments sit on the X chromosome, and they are adjacent and highly similar to each other.

That similarity makes them prone to recombination errors, which is why variation in red-green perception is comparatively common.

Someone with one X chromosome has no second copy to compensate, which is why the difference appears far more often in men than in women.

Plate tests exploit the confusion directly

The familiar coloured dot plates place a figure in colours that differ from the background along the axis a particular pigment difference collapses.

The dots vary in brightness and size deliberately, so the figure cannot be found by lightness or shape, only by the colour distinction being tested.

Some plates are designed so that a figure is visible only to people with the difference, which distinguishes genuine variation from guessing or from poor lighting.

Arrangement tests measure severity

Where a plate test detects that a difference exists, arrangement tests establish which type it is and how pronounced it is.

These require caps or chips of gradually changing hue to be placed in order, and the pattern of errors indicates the axis of confusion.

This detail matters where an occupation has colour requirements, since the standards usually specify a type and degree rather than a simple pass or fail.

Acquired changes are a separate concern

Inherited differences are present from birth, stable throughout life and affect both eyes identically.

A change in colour perception that develops later, or that differs between the eyes, points to something acquired rather than inherited.

That distinction matters clinically, and any noticed change in how colours appear is a reason to be examined rather than to assume an inherited variation was simply unnoticed.
